Stability of mAbs/ADCs

It's the best stability platform we use. We have 3 across departments and we use our uncles differently and we like how flexible it is. They have a lot of applications using the same hardware. One group uses it for thermal stability (Tms/Tagg and DLS). We do a lot of formulation screening for various programs and it's helpful to have had the same platform but also being able to do isothermal studies for long periods of time. The software is modern and light. It's fantastic.

This instrument has quickly become a standard method for a variety of applications including nanoparticle characterization, formulation optimization, and routine protein characterization. With minimal training, unskilled users can become proficient performing SOP methods. Expert users have developed custom methods that have advanced difficult projects. The instrument gives highly reproducible results, although the high sensitivity can make interpretation of some data features difficult. Standard DLS sample preparation considerations apply, and some care in loading samples must be taken. Some people love the software, some people dislike it, but in my opinion, most instrument software is imperfect anyway. Export functionality allows easy interfacing with custom analysis tools. We have had three service calls, all which were resolved quickly. The only downside is the high cost of consumables and some software related issues.

Screen proteins for structural biology

Our results using the Uncle are great. The DLS feature is very useful for seeing large aggregates and the relative size of particles after a very short time. The full spectrum fluorescence is also handy, especially when we are measuring the binding of a compound and want to see a shift in Tm. These two features work very well together. I would recommend it to any lab that requires screening of proteins very quickly (buffer screens, detergent screens etc.)
