Screening with the Yeast 2-Hybrid System on iQue

The Y2H screening system is a protein-fragment complementation assay that uses the power of molecular biology to screen for protein-protein (PPI) and protein-DNA (PDI) interactions. Traditional Y2H assays brought the bait and prey yeast strains together on solid agar that would turn blue when the LacZ reporter enzyme was expressed. The colonies expressing the reporter protein were picked off the surface and cultured. This process is effective, but the color development is slow and the colony picking is tedious. This application note describes how the Y2H system is readily adapted to the iQue by changing 2 aspects of the traditional system.
