CELL HANDLERTM, Cell picking and imaging system

The CELL HANDLER is an automated, damageless system for isolating a single cell or a single organoid/spheroid through imaging, picking and transfer of the cells individually to microtiter plates or PCR tubes, 3D-cells grown in hydrogel medium can also be isolated. This morphological/phenotypic image-based isolation system is applicable for various uses, e.g. cell-based assays, omics studies and cell line development.

Sort 3D structures (spheroids, organoids) for cell culture and subsequent handling

We are a service company (CRO) that does a lot of 3D cell culture work, and this instrument has dramatically changed our ability to work with 3D structures, generate high-quality and reproducible assays, and consistently select and sort structures for subsequent cell culture and fluorescence imaging work. The instrument is intuitive to learn, but we've found it most efficient to have dedicated personnel that are familiar with its operation. The availability of both bright-field and fluorescence options allow sorting of both live-cell structures (e.g. we've used it to build bright-field algorithms to select for 'healthy' 3D structures in subsequent assays) and tagged/labelled/fixed structures (e.g., for transfected/transduced structures).

The CELL HANDLER provides an automated cell isolation system consisted with image-based selection and subsequent picking and transfer of the selected cells individually to appropriate destination plates/tubes by pipetting-like manner. Either single cells or organoids/spheroids, not only cells grown in liquid medium but also those grown in semi-solid medium (e.g. Matrigel®), are applicable for this automated cell isolation system.

Briefly, morphological/phenotypic features of cells are obtained automatically by high throughput bright-field and fluorescent image analysis, so that users can easily select cells based on several cellular properties and histograms, namely “Criteria Selection”. Subsequently, the 8 individual tips are capable of picking and transferring the selected cells gently and rapidly, resulting in lowering cellular damage during plating/harvesting.

Various types of microtiter plates and microtubes can be chosen as destination depending on the following analysis including cell-based assays, omics studies and cell line development. In addition, custom plates such as Organ-on-a-Chip are also available.

(Key benefits)

  • Criteria selection - Automated identification and visualization of a single cell or a single organoid/spheroid of interest on the basis of cellular morphological/phenotypic characteristics
  • Improvement of data quality – Subsequent isolation of the selected cells individually and precisely results in improved uniformity of cell population and hence data quality
  • Direct harvest from the semi-solid medium - a single organoid/colony can be isolated without contamination of other adjacent ones
  • Improved success rate - Improvement of success rate of the isolation in comparison with limited dilution
  • Broad applicability - Standard and unique labware (e.g. Organ-on-a-chip)

(Key features)

  • Multiple imaging mode - microscope system equipped with 4x and 10x lenses is capable of bright field and 3-color fluorescence observation
  • Wide range head unit - precisely controlled 8-head and ɸ180-2200 μm tip lineup realize various picking and transfer
  • Sterile environment cabinet - cabinets with HEPA filters and UV lamps provide sterile condition for whole isolation procedure
  • Flexible image processing - specially designed image processing supports detection from single cell to organoid/spheroid
  • Traceable data output - all data (images, cell features, picking conditions) are output in traceable format for easy analysis

(Application examples)

  • Phenotypic screening
  • Antibody screening
  • Cell line development (Quality management, Transfection)
  • Single cell research
  • Stem cell research (iPSC, ES, Organoid)
