Lowered the barrier for and streamlined our stem cell gene editing workflow

Human iPSC clonal cell line generation

The IsoCell/IsoHub platform has streamlined the most finicky step of our human iPS cell line gene editing workflow. Rather than hand-picking of clones, we use the isoCell to automate most of the tedious and stressful (on the cell) steps from single cell to stem cell colony. Importantly, the small volumes in the isoCell have allowed us to truly start from single hiPSCs, rather than from clumps of cells or guessing whether we are making clonal cell lines. The automatic tracking with the IsoHub scope has made it easy for us to refer back at any step to quantitate and evaluate sample quality, even after our downstream quality control steps. IsoCell has sufficiently lowered the barrier to entry that now an undergraduate can be quickly trained to do clonal stem cell line generation for gene editing, which is important for us as an academic lab, and lets us create new gene-edited cell lines faster. The iotaSciences team was and continues to be great in providing hands-on training and support as we set up our methodologies, and is always open to feedback on how to make their product better fit our needs. Cloning Platform – THE single-cell cloning solution

The Cloning Platform consists of two core instruments – the isoCell and isoHub, with optional imaging and fluorescence capabilities available.

isoCell – so much more than a single-cell dispenser
The isoCell instrument automates all liquid-handling and minimises reagent wastage. It dispenses cells, exchanges culture media and harvests clonal cultures for downstream applications, all without the need for tedious manual pipetting. With the isoCell, cells are handled using low pressure and low shear forces. Together with optional heating, it delivers exceptional cell viability. Traditional single-cell cloning methods utilise volumes of cell culture media that quickly add up and rapidly increase the cost of your experiments; alternatively, the isoCell uses nanolitre culture volumes, drastically reducing costs and wastage.

isoHub - easily verify monoclonality and track your clones
Chambers can be easily viewed without any of the optical edge effects typically associated with conventional culture plasticware e.g. 96-well plates. Using our GRID technology, you have absolute confidence in identifying chambers that contain a single cell immediately after plating on day 0. The isoHub also automates navigation through chambers and allows tracking of your selected clones, using either using brightfield or fluorescent mode (if available). What’s more, data is wirelessly transferred between the isoCell and isoHub, guaranteeing a seamless and efficient workflow without the need to manually note single-cell chamber coordinates.

Image Capture System - document monoclonality with whole-chamber images
The optional Image Capture System lets you document monoclonality and clonal outgrowth in a single click using whole-chamber imaging. Simply select GRID chambers that contain a single cell, and the imaging system records and documents your selection with a whole-chamber image. Users can easily take follow-up images of relevant chambers throughout the cloning process to document clonal outgrowth. Moreover, image files are auto-named and sorted according to the chamber's details, providing a start-to-finish history of each single cell, for a fully transparent process record.

isoHub Fluorescence - 4 channels and 16 LED wavelengths to detect any fluorophore
The Cloning Platform optionally features a CoolLED pE-4000 illumination system for fluorescence microscopy. Easily detect commonly used fluorescent molecules (GFP, RFP, DAPI, and more) thanks to the system’s 4 channels, 16 LED wavelengths, and a user-friendly control pad. With simple one button on/off operation, it couldn’t be easier.
