Transfecting Agent
Although fugene 6 was ok to use it was discontinued and I had to switch to another reagent in middle of my study. The problem with fugene and other transfecting reagent is that it evaporates very easily and the container that it comes in is not made appropriately to protect it from evaporation. Also you can’t aliquote because of this same issue.

Plasmids Transfection
I have used fuGENE 6 for my transfection of plasmids. Their protocol is easy to use. A good point is that it is less toxic in the cell.

neuro2A Cell Line
It is working well in the cell line but not well in the primary neuronal culture. Especially for the div over 12.

Adenosine And Dopamine GPCRs Expression In Cell Culture
We used this to utilize lipofectamine to make stable transfection and transient transfections. We recently acquired FuGene and since then it has been easier and more reliable to make the transfections needed.

This works very well for neuronal cells transfection as well as an electroporation. It is easy use, quick and works well for primary cells.

It is a good reagent to work with in general. With some cell lines (U2OS) I have found it not working as well as in others (such as HeLa).

Cell transfections
Very good levels of transfections. Only requires a small volume- up to 6ul for a 10ml plate of 293 cells. Good for making retrovirus on 293 cells.

- Avoid time-consuming optimization procedures by relying on high transfection efficiency proven in more than 700 cell types, including many "hard-to-transfect" cells.
- Generate physiologically relevant results with the low-cytotoxicity of FuGENE® 6 Transfection Reagent. A recent gene-chip study demonstrated that a leading competitor´s transfection reagent significantly influenced the expression of more than 2,000 genes, compared to only 30 genes affected by FuGENE® 6 Transfection Reagent.
- Transfect in serum-free or serum-containing media. FuGENE® 6 Transfection Reagent is highly efficient both in the presence and absence of serum. In most cases, the same cell culture medium used for routine cell culture (e.g., one that contains serum) can also be used during transfection. This is especially useful in cases where transfection efficiency improves in the presence of serum.
- Save time and effort by eliminating multiple handling steps. Simply dilute FuGENE® 6 Transfection Reagent, incubate it with plasmid DNA, and pipet the mixture directly onto your cells (with or without serum).
