Cloning
I routinely use this for all DNA assemblies. At this point in my career, I have had other cloning methods fail too many times to bother using anything other than NEB Hifi. It works. It is robust.

Cloning
After learning cloning with standard restriction/ligation, I was introduced to Gibson assembly at the start of my postdoc. Using this, I've been able to clone large genes quite efficiently with up to 5 pieces to assemble. I would definitely recommend it to anyone. NEBuilder also has online tools to design your cloning and determine the amount of each part needed so tip top!

DNA Assembly Cloning
This kit has replaced "regular" cloning for me, at least if I need more than 2 fragments cloned into a backbone. Super-easy to use and takes very little time. In my experience, it works better when I use the upper limit of the suggested amounts for the individual fragments. Also, while it dosen't always work as expected, it literally is a hit or miss. Meaning either all clones are correct or none.
