Bacterial DNA purification from diverse samples containing PCR inhibitors: stool, soil, food etc.
PCR inhibitors and particulates are common within fecal and soil samples and can complicate downstream use (in my case, direct Sanger sequencing of PCR product). This kit (available in different capacities - I have used both microprep and miniprep kits) is designed to deal with these problems, by incorporating an initial column to remove particulates and a final column to remove PCR inhibitors. I have used this kit to successfully detect and type bacterial pathogens in stool. But beyond that, the kit has proven ideal for other "messy" samples, including food (poultry, etc.) rinses, food enrichments, and environnmental samples (specifically, listeria and c. diff cultures from environmental sampling tests that incorporate agar and other particulate or PCR inhibitor components). Indeed, several comparisons that I made of DNA preps from complete purifications versus purifications without the final inhibitor-removal column have demonstrated the importance of that final step. I routinely use this kit for DNA preps now, because with diverse samples you never know when an inhibitor might rear its ugly head!

Soil microbial metabarcoding
We have been users of the Zymo Soil DNA kits for years, first the ZR Soil Microbial DNA Microprep kit (D6003), then the Quick-DNA Fecal/Soil Microbe Microprep (D6012), and most recently the ZymoBIOMICS (Soil) DNA Microprep (D4301). All have been easy to use and consistent in their results. Indeed, when a previous PhD student started her work on metabarcoding from peat, I had her do a side-by-side comparison of the Zymo kit with kits from two other manufacturers, some of which were touted as especially designed for high humic acid samples. The DNAs recovered using the Zymo kits were more consistently PCR-amplifiable than those recovered using the other kits.
