rRNA depletion prior to RNA sequencing
We ordered riboPOOL for human samples after Illumina discontinued the RiboZero Gold standalone kit. We needed a rRNA depletion solution that worked not only for RNAseq (and we prefer enzyme-free kits) from total RNA but also for Riboseq on short (17-34 nt) ribosome protected fragments (most of the kits available in the market aren't compatible with this application). We first tested by RTqPCR the depletion of 18S rRNA from total human RNA (MDA-MB-231 cells) and got >2000-fold depletion with equal input of pre- and post-treatment samples. We then sequenced RNA from these samples and got 0.97% of total reads aligning to rRNA. We also tested riboPOOL performance in depleting rRNA fragments from ribosome protected fragment preparations for Riboseq and got 28.07% of reads aligning to rRNA (~2X more than with Illumina's RiboZero Gold in our previous experiments). Overall our experience has been very positive with riboPOOL, the tech support was prompt to reply and the cost of the product is very competitive.

Works great when you are doing RNA sequencing. riboPOOL kit is species specific. I would highly recommend using this kit. It's user friendly, economic and is sensitive and specific.
