Great instrument for transfections.

After struggling with transfection of human embryonic stem cells using other transfection methods, the Lonza 4D nucleofector was a great choice for me. Lonza's 4D nucleofector system is easy to used and easy to optimize the sgRNA concentration, no. of cells and program using the 16 strips cuvette. You can choose different programs in each 16 strips cuvette and it only takes a few seconds for nucleofection. I got high transfection efficiency with high viability. This system is much better than liposome based transfections and other transfection system.

Electroporation of neuronal primary cultures

The Nucleofector is very easy to use but give quite low levels of expression of your construct of interest, at least in neuronal rat primary cultures. Moreover, a very high number of neurons are needed due to the high mortality of the electroporation procedure. In addition, you must use the manufacturer solutions for the procedure, with homemade solutions mortality increases and efficacy decreases. I know there is a new version of the instrument now available and they may ensure better results.

Our Nuclefector is used to transfect a wide range of cells, ranging from primary cells to cell lines. Nice efficiency for siRNA transfection into both primary cells and cell lines. On the other hand, if electroporation of DNA plasmids is efficient for cells lines, it is rather limited for primary cells (especially neurons and dendritic cells). A bit expensive, but really user friendly.

I use the Nuclefector Kit L and have found it to be simple and practical. It is the primary culture of rat cortical neurons. However it is expensive when you have to use for large scale.

The Nucleofector technology is a highly efficient non-viral method for transfection. It is based on two components: the Nucleofector Device that delivers unique electrical parameters, and Nucleofector Kits that contain cell-type-specific Nucleofector Solutions.

The electrical parameters of the Nucleofector are different from any other commercially available electroporation device. Each electrical setting is displayed as a distinct program which has been adapted to the requirements of a particular cell type. As the electrical settings are pre-programmed into the Nucleofector, optimization of the electrical parameters by the user is not necessary.

The device weighs only 2.6 kg and is conveniently sized so that it easily fits under a sterile hood, which greatly facilitates sterile work with cells.
