Purification of fungal genomic DNA for DNA barcoding identification

This was an easy to use product for rapid purification of genomic DNA from a variety of fungal samples. We had originally used the Promega Wizard kit but it was time-intensive and did not yield sufficient quantities of clean genomic DNA. We were able to improve our PCR efficiency with DNA obtained from fungi using this kit. Also, we were able to teach undergraduates students how to purify DNA in rapid manner.

This product yielded excellent DNA extractions from Saccharomyces cerevisiae mutants even after multiple failed attempts to use other protocols (non-kit) to obtain DNA from the same strains. These extractions were then able to be used successfully in PCR!

We have been using this kit for 2 years now. We have used it for purification of genomic DNA from a wide range of fungal species with a high rate of success. Purified DNA is used as a template for DNA barcoding identification of fungal species. We have seen a much higher rate of successful PCR from this kit than other commercially available kits.

The YeaStar™ Genomic DNA Kit is designed for reliable and efficient isolation of genomic DNA from a broad spectrum of fungal species, including Aspergillus fumigatus, Aspergillus nidulans, Aspergillus nivens var. aureus, Candida albicans, Pichia pastoris, Saccharomyces cerevisiae, Schizosaccharomyces pombe, and any fungi whose cell walls are susceptible to yeast lytic enzyme lysis. The kit is based on highly efficient enzyme lysis and Zymo-Spin™ column technology. Each standard prep yields about 7 - 20 µg of DNA with a size distribution of 35 - 60 kb. The resulting genomic DNA can be used direct analysis including Southern blotting, PCR, restriction endonuclease digestion, etc.
