Plasmid purification for mutagenesis and transfection
Cell Source: Thermo TOP10 Cells Culture Volume: 100mL Process Time: 8 samples takes ~2-3 hours from before culture spin down to completion of elution The protocol says 50mL of culture but we have adapted it for use with 100mL. The column say that they can hold up to 1.2mg of DNA and this culture volume works for us. This protocol is very resilient to user error and is consistent enough that we have had new interns reaching competency very quickly on this protocol. We have been using this product for over 1 year and the failure rate (<100ug) is <5%.

Easy to use & adjust to experiments
Bacmid DNA Purification
This Zymo kit is a breeze to use- the instructions are clear, visual, and detailed, and the color-coded reagents make it easy to identify at a glance what is what. The kit is also readily adapted to the specifics of the experiment. If increasing the DNA yield or removing endotoxins is important, there are optional steps to include in your procedure. Also, replacing individual kit components does not require you to buy a whole new kit- every component has a part number that allows you to buy it directly from the manufacturer's website. Would highly recommend this product!

Plasmid Extraction
When I first started my Ph.D., I had to do a lot of plasmid extractions. I struggled for months using different kits. I convinced my PI to purchase the Zymo Midiprep kit and I have successfully extracted high-quality plasmids without fail. Now, I can finally move on with my research and not wonder if I will get a high yield of high-quality plasmid. One of my favorite things about this kit is that the buffers are different colors. This is especially helpful for the neutralization step which allowed me to be confident in moving on to the next step. Another thing that I love about this kit is that the elution volume is 200uL. Other kits elute 1mL from the column which means that you need to precipitate the DNA in order to get a high concentration. This was especially important for me since I regularly needed 1ug/uL of plasmid DNA. I love everything about this kit and it is very quick to use. One of the best things is that it's cheap and of VERY good quality! Now, my lab has transitioned to using Zymo for Miniprep, Midiprep, and Maxiprep! Thank you, Zymo!!!

ZymoPURE ™ Plasmid Isolation kits provide the fastest and simplest method available to efficiently isolate transfection quality plasmid DNA from E. coli. The ZymoPURE ™ Plasmid Midiprep kit features a spin column-based method for the purification of up to 300 µg of high-quality plasmid DNA in less than 18 minutes. The eluted plasmid DNA is ready for immediate use, avoiding the need for subsequent precipitation steps.
ZymoPURE ™ technology uses a modified alkaline lysis method and features novel binding chemistry that yields highly concentrated plasmid DNA (up to 3 µg/µl) directly from a spin column. The wash regimen has been optimized to ensure the plasmid DNA is free of endotoxins, salt, protein, and RNA resulting in plasmid DNA suitable for sensitive applications including transfection, plasmid-mediated gene silencing, cloning, and sequencing.
As an added convenience, the ZymoPURE ™ Plasmid Midiprep contains colored buffers that permit error-free visualization and identification of complete bacterial cell lysis and neutralization. Syringe filters are included for rapid clearing of the lysate and the unique spin-column design allows the binding step to be performed using a vacuum or table top centrifuge.
