I use the Zymo mini-prep for plasmid DNA isolation and it is the easiest prep I've worked with. All of the components needed along with the tubes and spin columns are included in the kit. It's like making premium brownies in your own kitchen. The prep always works, the DNA is clean enough to do anything I want with it and it always turns out great. The yield is good and the prep is quick... depending on how many samples I am trying to make, I spend under two hours isolating my DNA.

Plasmid purification for mutagenesis and transfection
The whole protocol from before spinning down culture to final elution is ~2.5 hours for 20 samples. Cell Source: Thermo TOP10 cells Culture Volume: 5mL Very easy to use, there is a lot of flexibility in timing of steps if something comes up while processing. The methodology is well thought out and resilient to user error. Product states that one can expect 100ug, but from user experience when the process is handled with care we have been able to purify ~200ug from 5mL overnight culture. As long as the cells have no problems with growth, 75-100ug is a reliable target that should be reached with every prep. User failure rate for our group (<20ug) is <5%.

Great results from this product. Provides high yields and transfection-grade plasmid DNA using spin columns. Recent protocol update no longer requires one of the reagents to be ice cold which saves time.

Plasmid DNA extraction
I have purchased this large kit 3 times already. I have found it is easy to use with clear protocol instructions. However, in this last kit I noted many columns were missing the purple insert, and sometimes the column filter would float upon addition of buffer. I also noted they do not add 10% extra to some of their buffers and the kit comes with exact volumes for the reactions. Note this is the case with the Wash 1 buffer. I am surprised given most companies will include extra solutions to account for pipetting errors. Not sure I will keep buying the kit if these two things are not fixed. Value for the money is great!
