Method Development
In HPLC/UPLC, methods in which mobile phase is run in the form of a binary gradient, it is seen in many cases that ghost peaks appear in the baseline which interfere with analytes and sometimes with main peak. These arise due to impurities in solvents i.e. mobile phase. It is suspected by scientist that it is because the needle is not clean. However, while developing a method I saw that a ghost peak appeared on some columns exactly at the retention time of my main peak. Surprisingly, this did not happen with one of the columns which confirmed that the ghost peak was not related to needle contamination. However, after attaching a separate ghost column between pump and the injector (your injected sample does not pass through it but only mobile phase passes through it) the ghost peak simply disappeared. Moreover, the humps in the mobile phase got greatly reduced making the peaks of interest quantifiable with ease. In the past, I faced this problem of ghost peaks appearing at the same retention time as my main peak, impacting the area of diluted standard in the impurity method and also putting a question mark on the specificity of the method. But after using ghost column, this problem is totally solved! It would have been better had I come to know this solution earlier. However, this technology is recent development. An amazing product!
