In the case of GST or 6HIS tagged proteins, this verification can be achieved with HTRF® GST and 6HIS check kits. These two assays are competitive immunoassays in which the anti-GST or anti-6HIS Cryptate conjugates bind directly to XL665-conjugated GST or 6HIS peptide.
A signal decrease proportional to the amount of tagged protein to be tested is the sign of accessibility of the tagged moiety for the corresponding HTRF® toolbox reagents (anti-GST or anti-6HIS).
Each kit yields 1,000 test determinations using a 20 µL final volume. Each kit contains lyophilized anti-GST or anti 6HIS Cryptate conjugates, and GST-XL665 or 6HIS-XL665 conjugates.
It also includes a maximum calibrator for the constitution of the calibration curve (GST check kit) or a kit control (for 6HIS check kit). Reconstitution buffer and diluent are supplied for both kits.
