When I use prime HLB, there are certain non polar compounds that can not retain on the cartridge but can retain well on C18. And also prime HLB has less loading amount comparing to HLB. However, prime HLB is easier to use.

Plant extract purification
The purification protocol through OASIS HLB prime cartridges is well described in Waters website and in different experiments carried out by Waters scientists. I have to say that the use of these cartridges is very user-friendly, especially when coupled with the Waters Extraction Manifold system. The main advantage is the avoiding of the first two steps of activation and equilibration of the cartridge, that results in saving at least 10-20 minutes. This can be achieved since the matrix of the cartridge is water wettable (HLB resin presents both lipophilic and hydrophilic retention features). The purification could be made by following one of the two possible procedures and the decision depends on what the user is looking for (i.e. purification from sugars? Purification from phospholipids?...) and the extraction solvent that has been used to extract the metabolites (water, buffer, or methanol). The first strategy (A) occurs when the extracts consist of water or buffer. In this case, sugars and polar metabolites are removed from the sample. The extracts must be passed through the cartridge without any previous operation, then water can be used to wash the cartridge from sugars and other very polar metabolites. This step can be repeated twice. Subsequently, methanol or other less polar solvent can be used for the elution of mildly, less polar metabolites. b) In the other case, methanolic extracts must be supplied with acetonitrile to reach the final ratio of 10:90 of Methanol:Acetonitrile (v/v). Then, the solution can be passed through the cartridge and the flow trough can be collected. The main disadvantage of this point is that you will have a more diluted sample, but phospholipids and other contaminants will remain attached to the cartridge, thus reducing the matrix effects in the following mass spectrometer analysis. Moreover, one can overcome the dilution problem by using a speedvac system to evaporate the solvent and then re-suspend the resulting “powder” with methanol. However, special attention should be taken into account since some metabolites could degrade during the evaporation process. We are still evaluating this point for our analysis. In summary, Oasis Prime HLB might be a precious tool for sample purification, especially from those metabolites which can affect the ionization in mass spectrometry, such as salts, organic acids, sugars and phospholipids. They are also a flexible tool since prime HLB can be used before or after other Oasis cartridges, for example MCX or MAX. I would recommend the use of these cartridges for quantitative analysis in targeted and untargeted metabolomics.

Solid and liquid extraction in forensic analysis
We use the 30mg MCX cartridges, but they are very expensive. Also 96 well plates are not very interesting for microelution because sometimes we can not use all the plates and it is difficult to put a label on each column to avoid getting too much sample.
