DNA extractions and PCR amplification for downstream sanger sequencing
I use this product routinely to verify CRISPR knockouts generated in the laboratory. This solution makes it remarkably easy to extract genomic DNA from either cell pellets or attached cells. I was able to quickly harvest gDNA from 48 clones directly from 24 well dishes just by aspirating media, washing in DPBS, and adding the QuickExtract solution directly to the wells. After harvesting, a 15 minute incubation protocol will have the gDNA ready for PCR. Very concentrated gDNA can interfere with downstream PCR reactions, but Lucigen sells a polymerase kit optimized for amplifying samples in QuickExtract solution, or you could just add more QuickExtract solution to dilute your gDNA (I repeat incubation after diluting).
